DNA hybridization testing

Preapre a solution consisting of 0.5 M NaCl, 0.01% Tween 20, and 10 mM PBS. Put probe and target DNAs (1:20) into the solution.

Place a droplet of solution on a substrate holding capture DNAs and incubate the substrate:
1) for 15 min at 60°C
2) for 30 min at 37°C
3) more than 2 hours at RT

Testing procedures

  • Six droplets were placed on a gold substrate.
  • The substrate was kept in a 50% humidity chamber for 20 min.
  • The substrate was rinsed with water and passivated with C6 thiol by immering the substrate in the solution (1 mM C6 in ethanol) for 5 min.
